While handling my Chilean Rose-Haired Tarantula, it started spinning a web around my hands. Should I be worried? Was she stressed?

No, you shouldn't be worried. Tarantulas use webs/silk to line their underground
burrows or where they hang out in the cage. They can also use silk to help with
traction when climbing objects. Perhaps the spider was simply trying to
increase its traction while climbing around on your hands.
