There are lots of factors including what variety of watermelon you are growing. However, most pollen degrades quickly once it is out of contact with the source plant. Labs that do experiments saving pollen for whatever reason have equipment and an environment you can't duplicate at home. That said, you can try various experiments just to find out for yourself. You can try saving different male flowers in the refrigerator, the freezer, by drying, etc and see what happens. A nice moment of science.
