What could be tunneling inside the tomato leaf vein and causing cupping of the tomato leaves? I don’t see any signs of aphids, nor spider mites. I don’t see any insects at all on the plant. Nearby plants are not showing symptoms yet. I’ve been applying an organic pest spray.

You may have a couple of things going on. Leaf miners cause the tunneling and there is a caterpillar on the top leaf. If there are only one leaf affected, remove it and dispose of it.
the caterpillar can be removed by hand. You may have to get a stronger spray.
You may have to get a stronger pesticide to rid the leaf miners. You can google for suggestions as I am not allowed to suggest specific brands. Keep an eye out for more .
